چکیده

Objective: Ricin is a highly toxic ribosome-inactivating lectin derived from castor beans. To date, no antidote available to treat ricin-poisoned patients, and the development of safe effective urgently needed. Methods: First, ricin was prepared used construct mouse model rhesus monkey intoxication. Second, pepsin-digested F(ab′) 2 fragments serum IgG horses injected with Freund’s-adjuvanted purified were prepared. Third, protective efficacy evaluated in models lethal Results: The purity quotient exceeded 90% 85% models, respectively. LD 50 mice monkeys 2.7 9 μg/kg, A quantity 6.25 1.85 mg/kg sufficient intoxication monkeys, Finally, effect this therapeutic antibody on peripheral blood immune cells examined by analysis through single cell sequencing. underlying mechanism found involve restraining neutrophil activation, proliferation, differentiation. Conclusion: Purified administered needle-free devices fully protect against doses

Immunoglobulin A antibodies against ricin A and B subunits protect epithelial cells from ricin intoxication.

Epithelial cells of the respiratory and gastrointestinal tracts are extremely vulnerable to the cytotoxic effects of ricin, a Shiga-like toxin with ribosome-inactivating properties. While mucosal immunity to ricin correlates with secretory immunoglobulin A (IgA) antibody levels in vivo, the potential of IgA to protect epithelial cells from ricin in vitro has not been examined due to the unavail...

Inhibition of Retrograde Transport Protects Mice from Lethal Ricin Challenge

Bacterial Shiga-like toxins are virulence factors that constitute a significant public health threat worldwide, and the plant toxin ricin is a potential bioterror weapon. To gain access to their cytosolic target, ribosomal RNA, these toxins follow the retrograde transport route from the plasma membrane to the endoplasmic reticulum, via endosomes and the Golgi apparatus. Here, we used high-throu...

A Single Phosphorodiamidate Morpholino Oligomer Targeting VP24 Protects Rhesus Monkeys against Lethal Ebola Virus Infection

UNLABELLED Ebola viruses (EBOV) cause severe disease in humans and nonhuman primates with high mortality rates and continue to emerge in new geographic locations, including several countries in West Africa, the site of a large ongoing outbreak. Phosphorodiamidate morpholino oligomers (PMOs) are synthetic antisense molecules that are able to target mRNAs in a sequence-specific fashion and suppre...

Thermostable ricin vaccine protects rhesus macaques against aerosolized ricin: Epitope-specific neutralizing antibodies correlate with protection.

Ricin toxin (RT) is the second most lethal toxin known; it has been designated by the CDC as a select agent. RT is made by the castor bean plant; an estimated 50,000 tons of RT are produced annually as a by-product of castor oil. RT has two subunits, a ribotoxic A chain (RTA) and galactose-binding B chain (RTB). RT binds to all mammalian cells and once internalized, a single RTA catalytically i...

Purified influenza virus nucleoprotein protects mice from lethal infection.

Local administration of nucleoprotein purified from X31 (H3N2) influenza A virus primed for A virus cross-reactive cytotoxic T cells and resulted in substantial protection (75%) of mice from a lethal challenge with the heterologous mouse-adapted A/PR/8/34 (H1N1) virus. By following the course of a lethal virus challenge we found that nucleoprotein priming did not prevent virus infection but rat...
